Wettability-Gradient Multilayer Nanofibrous Membrane with Unidirectional Self-Pumping Capability for Wound Dressings

  Abstract

Chronic wounds, especially diabetic ulcers, often produce significant exudate during healing. While conventional dressings can temporarily replace damaged skin and offer a protective barrier, excessive exudate accumulation may extend inflammation, increase infection risk, and delay wound healing and regeneration. In this study, we developed a novel multilayer fibrous dressing featuring a Janus-structured wettability gradient using layer-by-layer electrospinning technology. The composite structure comprises: (1) a skin-facing layer with hydrophobic PCL-PVA/CS and hydrophilic PVA/CS/AMOX components; (2) an intermediate PCL/Gel layer for efficient fluid absorption and retention; and (3) an outer medical non-woven fabric layer. The resulting multilayer fibrous membranes demonstrate exceptional unidirectional fluid transport, allowing complete permeation of simulated exudate across the dressing within 24 s through their self-pumping effect. When loaded with amoxicillin sodium, the membrane shows strong antibacterial activity against both Staphylococcus aureus (S. aureus) and Escherichia coli (E. coli). Additionally, comprehensive testing reveals superior performance in moisture management, mechanical strength, biocompatibility, drug loading and release kinetics, and resistance to protein adsorption. These combined properties make the wettability-gradient multilayer fibrous dressing a highly promising solution for advanced wound care applications.
